Pharmaceuticals and Drugs

Nowadays, many types of pharmaceuticals are prescribed for the treatment of various diseases. For sensitive analytical determinations of pharmaceuticals from body fluids efficient methods using SPE and HPLC mostly followed by MS/MS detection are necessary.

The new hydrophilic-lipophilic balanced polymeric SPE adsorbent CHROMABOND® HLB is specially designed for the enrichment of hydrophilic analytes. Applications for sulpha drugs, tetracyclines, and iodinated contrast media as well as for mixtures of pharmaceuticals were successfully developed (see www.mn-net.com/apps).

For the subsequent HPLC analysis the professional porous silica phases NUCLEODUR® and the highly efficient core-shell phases NUCLEOSHELL® are used. The hydrophobic C18 phase NUCLEODUR® C18 Gravity-SB with its distinct polar selectivity shows good results for most pharmaceuticals - also in accordance with Pharmacopoeia (e.g., cefepime dihydrochloride, betamethasone dipropionate). With the biphenylpropyl phase NUCLEODUR® π2 steroids, tetracyclines, and further veterinary drugs can be very well separated. The latest development of core-shell phases NUCLEOSHELL® Bluebird RP 18 - an octadecyl modification with polar endcapping - allows fast analyses for e.g., antibiotics, and radiocontrast agents as well as for drugs of abuse like THC, and amphetamines.

TLC with e.g., SIL HD and HPTLC with nano-SIL HD glass plates are very suitable for the analysis of natural plant compounds for biopharmaceuticals due to the advantages against matrix effects, compared to HPLC methods.

Food and Beverages

MACHEREY-NAGEL offers ready-to-use QuEChERS mixes according to EN 15662 and AOAC 2007.01 standards. Applications for the extraction of many pesticides from fruits and vegetables, acrylamide in coffee, and dyes from aquaculture samples can be found at www.mn-net.com/apps. For complex extractions the SPE phase CHROMABOND® HLB and the concept of CHROMABOND® HR-Xpert are successful. The hydrophobic HR-X phase can be used for extraction of e.g., chloramphenicol in prawns or biocides in detergents and cosmetics. Whereby the mixed-mode cation exchanger HR-XC can be applied to the extraction of melamine in milk.

Additionally NUCLEODUR® and NUCLEOSHELL® HPLC columns are utilised for many applications (e.g., vitamins in supplements or juices, sweeteners in energy drinks, pesticides in food). For the separation of sugars the polymeric NUCLEOGEL® SUGAR columns are offered.

As food matrices are very complex TLC methods (e.g., analysis of vitamins or aflatoxins on SIL HD plates, characterised by high luminosity) are widespread.

Environmental Samples and Pollutants

The US Environmental Protection Agency (EPA) focuses mainly on urban and industrial pollution, water quality and electronic waste. EPA regulations are legal in many countries of the world.

For example, 16 EPA PAHs are separated on the special HPLC phase NUCLEODUR® C18 PAH. According to EPA 531.1 carbamate pesticides are determined on an NUCLEODUR® C18 Gravity-SB column. The broad selectivity range of this RP phase allows a good separation of different pesticide classes included their metabolites.

Polymeric SPE phases are very important for the analysis of pesticides from water. A lot of applications with CHROMABOND® HR-X have been established in many laboratories for water analysis. CHROMABOND® HLB can also successfully be applied for pesticides as well as for drugs from tap water. An enrichment of acrylamide from water according to DIN EN ISO 38413-6 using CHROMABOND® Carbon A is described in the selected MN application note 5/2017 at www.mn-net.com/apps.
