DUETTA™: Fluorescence and Absorbance in ONE Benchtop Instrument

Jul 04 2018

Duetta™ is a revolutionary fluorescence/UV-Vis spectrometer that combines fluorescence and absorbance spectroscopy into ONE instrument. It guarantees performance and accuracy for both measurements using the same experimental conditions. In addition, this feature provides a significant economy for laboratories by having the equivalent of two systems for the price on one.

The design of Duetta™ is focused on the ease of use and the quality of the obtained data. Thereby, Duetta™ includes an automatic inner filter effect corrections method. This useful feature is unique to HORIBA spectrofluorometers. Over an absorbance of 0.1, the fluorescence emission of the excited molecules can be reabsorbed by neighboring molecules. What is seen is a loss of the linearity rule between the intensity of the fluorescence signal and the sample concentration. In the best case, to avoid reabsorption, the user measures the absorbance to make sure it is below 0.1 and, in case of higher values, applies dilutions to the samples. This takes time and may introduce some artefacts. In the worst case, the user does not consider the reabsorption effect and obtains data than can be erroneous. With Duetta™, the inner filter effect is automatically corrected, meaning that there is no need for absorbance measurements and dilutions. Therefore, Duetta™ guarantees the accuracy of the measurements for all users.

Duetta™ is not only accurate. It is also extremely fast. Its ultra-fast, state-of-the-art CCD technology allows it to outperform, by orders of magnitude, any competitor using PMTs, making it the fastest bench-top fluorometer in the world! This state-of-the-art CCD technology also extends NIR wavelength detection to 1,100 nm, well beyond the limit of a standard PMT fluorometer. Duetta's use of a CCD lets you collect the entire emission spectrum in 50 milliseconds. This is a huge differentiator between Duetta™ and a traditional scanning spectrofluorometer. For more details, read the first article in our new 'Science in Action' series.

Duetta™ is ideal for molecular fingerprinting. In addition to traditional fluorescence and UV-Vis-NIR absorbance spectroscopy modalities, Duetta™ also features Absorbance-Transmittance fluorescence Excitation Emission Matrices (A-TEEM) measurements providing extended linear dynamic range due to automatic inner filter effect corrections.

Since its recent introduction in March 2018, Duetta™ has already been awarded by prestigious institutions:

  • The Pittcon Today Excellence Awards, Gold
  • Silver award from the Laser Focus World 2018 Innovators Awards Program

Along with its technical performance, Duetta™ is also being recognised for the quality of its new touchscreen software EzSpec™. In just a few minutes with EzSpec™, users will appreciate the care that has been applied to the ergonomics and ease of use of the software during its development.
