Defined hydrogel enables reliable cancer cell culture in xenograft models

AMSBIO has introduced MatriMix for PDX, a fully defined extracellular matrix (ECM) hydrogel specifically formulated to support patient-derived xenograft (PDX) studies. Comprising collagen type I, laminin-511 E8 fragments, and hyaluronic acid, this ready-to-use 3D culture substrate provides an optimised environment for culturing patient-derived cancer cells.

PDX models utilise tumour tissue from patients to establish tumours in immunodeficient mice, preserving the histological and genetic characteristics of the original tumour. These models offer an advanced platform for cancer research, enabling drug screening and toxicology studies to facilitate the development of novel treatments.

MatriMix for PDX is designed to deliver high lot-to-lot consistency, ensuring reproducible results for researchers. Its composition has been carefully optimised to create an ideal extracellular environment, supporting even challenging-to-culture cancer cell types in xenograft applications.

The hydrogel is made with medical-grade porcine collagen, facilitating a smooth transition from basic to clinical research, setting it apart from traditional natural hydrogels. Supplied frozen, MatriMix for PDX forms a clear gel when warmed to physiological temperature (37⁰C), enabling enhanced visualisation for real-time monitoring of tumour growth, invasion, and drug response assays.

MatriMix for PDX is available in a convenient, single-vial format, as well as a tunable three-vial version, allowing users to customise conditions for specific 3D cell culture or organoid models.
